Gynazole (Butoconazole) for vaginal candidiasis

Gynazole or Butoconazole inhibits ergosterol synthesis which is an important component of the cell membranes of fungi. It is used for the local treatment of vulvovaginal candidiasis due to Candida albicans

Butoconazole Dose in Adults

Use in the treatment of Vulvovaginal candidiasis:

    • Insert 1 applicatorful (equivalent to 5 gms) intravaginally as a single dose.

Butoconazole Dose in Childrens

Not applicable. 

Pregnancy Risk Factor C

  • Use of it in animal studies has been linked to adverse effects on fetal health.
  • The observed fetal effects may be caused by small amounts of drug systemically absorbed.
  • It is not recommended to take a single dose of therapy. Pregnant women should receive 7-day therapy for vulvovaginal candidiasis.
  • It can cause rubber condoms, diaphragms, or latex condoms to become less durable.

Butoconazole use during breastfeeding:

  • Breastfeeding women should not use it as the excretion into breastmik is unknown.

Butoconazole Dose in Renal Disease:

Dose adjustment has nt been recommended by the manufacturer in patients with renal diseases.

Butoconazole Dose in Liver Disease:

Dose adjustment has nt been recommended by the manufacturer in patients with liver diseases.

Side effects of gynazole:

  • Gastrointestinal:
    • Abdominal cramps
    • Abdominal pain
  • Genitourinary:
    • Pelvic pain
    • Vulvovaginal burning
    • Vulvovaginal soreness
    • Vulvovaginal pruritus
  • Local:
    • Local swelling

Contraindication to Butoconazole include:

  • Allergy reactions to butoconazole and any component of the formulation

Warnings and Precautions

  • It may contain mineral oil, which can cause rubber to become brittle or diaphragms to be ruined. These products should not be used by patients for more than 72 hours.

How to administer Gynazole (Butoconazole)?

  • It is used as an intravaginal cream. It is applied locally via the applicator provided by the manufacturer.
  • It should be inserted deep into the vagina (without causing discomfort).

Mechanism of action of Gynazole (Butoconazole)

  • It blocks the biosynthesis ergosterol, an important component of cell membranes in fungi.
  • This causes a rise in the permeability of fungal cells and damages them, resulting in death.
