Nedocromil (Alocril) Eye Drops - Uses, Dose, MOA, Brands

Nedocromil (Alocril) inhibits the release of inflammatory mediators from cells including mast cells, eosinophils, neutrophils, and macrophages. It is used to treat patients with allergic conjunctivitis.

Nedocromil Uses:

  • Allergic conjunctivitis:

    • Used for the treatment of itching associated with allergic conjunctivitis

Nedocromil (Alocril) Dose in Adults

Nedocromil (Alocril) eye drops dose in the treatment of Allergic conjunctivitis:

  • Ophthalmic: One to two drops in each eye two times a day throughout the period of allergen exposure.

Nedocromil (Alocril) Dose in Childrens

Nedocromil (Alocril) eye drops dose in the treatment of Allergic conjunctivitis:

  • Children ≥3 years and Adolescents:

    • Ophthalmic: One to two drops in each eye two times a day throughout the period of allergen exposure.

Pregnancy Risk Category: B

  • Pregnant women are not subject to well-controlled studies.
  • Animal studies have not shown any evidence of teratogenicity, or harm to the foetus.
  • It is very low in systemic absorption.

Use during breastfeeding:

 

  • It is unknown if the drug is excreted in breast milk.
  • The manufacturer recommends that nursing mothers be cautious when administering nedocromil.

Dose in Kidney Disease:

  • No dosage adjustments have been provided in the manufacturer's labeling.
  • However, due to low absorption in systemic circulation dosage adjustment is unlikely.

How to administer Nedocromil (Alocril)?

  • Only for ophthalmic use.
  • To avoid bacterial contamination, the tip of the container should not be touched to the eye, surrounding structures, fingers, or other surfaces.
  • During periods of symptomatic allergic conjunctivitis, the use of contact lenses is not recommended.

Mechanism of action of Nedocromil (Alocril):

  • This causes the inhibition of activation and release mediators from various inflammatory cell types that are associated with hypersensitivity reactions, including macrophages and monocytes, eosinophils and neutrophils, as well as inhibition of the release and activity of histamine, leukotrienes and slow-reacting substances of anaphylaxis.

Absorption:

  • 70% urine (as an unchanged drug)
